چکیده
: Metastasis spreading confers a worse prognosis to the clinical outcome among patients suffering from osteosarcoma, most common primary bone tumor in childhood and adolescence. The detection of molecules involved metastasis might contribute understanding dissemination mechanisms, thereby opening way novel therapeutic strategies. Ezrin-radixin-moesin (ERM) family proteins are activated after interacting with belonging phosphoinositide signal transduction pathway. phosphatydil inositol (4,5) bisphosphate (PIP2), crucial molecule PI pathway, stabilizes ERM or more efficient receptor binding. PIP2 levels pathway critical element for regulating several cell events. regulated using enzymes, including PI-specific Phospholipase C family. A decrease induces dissociation protein membrane. In this regard, PI-PLC enzymes regulate event. paper, role osteosarcoma metastases is discussed.

Silencing of phosphoinositide-specific phospholipase C ε remodulates the expression of the phosphoinositide signal transduction pathway in human osteosarcoma cell lines.
BACKGROUND Ezrin, a member of the ezrin-radixin-moesin family, is involved in the metastatic spread of osteosarcoma. Ezrin binds phosphatydil inositol-4,5-bisphosphate (PIP2), a crucial molecule of the phosphoinositide signal transduction pathway. PIP2 levels are regulated by phosphoinositide-specific phospholipase C (PI-PLC) enzymes. PI-PLCε isoform, a well-characterized direct effector of rat...
متن کاملberberine induced apoptosis of human osteosarcoma cells by inhibiting phosphoinositide 3 kinase/protein kinase b (pi3k/akt) signal pathway activation
background: osteosarcoma is a malignant tumor with high mortality but effective therapy has not yet been developed. berberine, an isoquinoline alkaloid component in several chinese herbs including huanglian, has been shown to induce growth inhibition and the apoptosis of certain cancer cells. the aim of this study was to determine the role of berberine on human osteosarcoma cell lines u2os and ...

The phosphoinositide 3-kinase (PI3K)/Akt pathway controls a vast array of normal physiological processes and is frequently aberrantly activated in cancer, thus identifying PI3K/Akt-signaling components as promising drug targets in oncology. However, implementation of rational cancer therapies for this pathway needs robust and simple tools to stratify patients according to PI3K pathway activatio...
متن کاملSignal transduction and membrane traffic: The PITP/phosphoinositide connection
Phosphatidylinositol (Ptdlns) transfer protein (PITP) is a ubiquitous and abundant cytosolic protein that was discovered some 25 years ago because it can exchange Ptdlns and phosphatidylcholine (PtdCho) between lipid bilayers (for review see Wirtz, 1991). This in vitro activity suggested an in vivo function of PITP in transport of these phospholipids from the site of their synthesis in the endo...
متن کاملSignal transduction pathway of TonB-dependent transporters.
Transcription of the ferric citrate import system is regulated by ferric citrate binding to the outer membrane transporter FecA. A signal indicating transporter occupancy is relayed across the outer membrane to energy-transducing and regulatory proteins embedded in the cytoplasmic membrane. Because transcriptional activation is not coupled to ferric citrate import, an allosteric mechanism under...
